Overview

Luconosatuzumab (also known as LNC1004) is a humanized monoclonal antibody designed to target **fibroblast activation protein alpha (FAP-α)**, a cell surface glycoprotein that is highly expressed on cancer-associated fibroblasts (CAFs) within the tumor microenvironment (TME). FAP-α plays a critical role in tumor progression, stroma remodeling, and immunosuppression, making it an attractive target for modulating the TME to enhance anti-tumor immunity. Luconosatuzumab is being developed primarily for the treatment of advanced solid tumors, including **gastric adenocarcinoma** and **gastroesophageal junction (GEJ) adenocarcinoma**. In clinical settings, it is being evaluated in combination with immune checkpoint inhibitors like toripalimab to overcome resistance to first-line immunotherapy. Additionally, the antibody serves as the targeting component for radiopharmaceutical derivatives, such as 177Lu-luconosatuzumab, which deliver targeted radiotherapy to FAP-expressing stroma.
