Lucorafusp alfa (AK130) is a first-in-class bifunctional antibody fusion protein developed by Akeso Inc. It consists of an anti-TIGIT monoclonal antibody fused to the extracellular domain of human transforming growth factor-β receptor II (TGFβ-RII). This dual-targeting approach aims to overcome immune evasion in the tumor microenvironment by simultaneously blocking the TIGIT-CD155 inhibitory pathway (reinvigorating CD8+ T cells and NK cells) and neutralizing TGF-β signaling (reducing immunosuppression and fibrosis). It is currently being evaluated in clinical trials for advanced solid tumors, including hepatocellular carcinoma and pancreatic cancer, both as a monotherapy and in combination with other immunotherapies like ivonescimab.
