Drug intelligence / Profile preview
Lumateperone LAI is a long-acting injectable formulation of lumateperone, an atypical antipsychotic developed for the treatment of schizophrenia and potentially other neuropsychiatric disorders. Unlike the oral capsule form (marketed as Caplyta), this formulation is designed for once-monthly subcutaneous administration to maintain therapeutic plasma levels over at least 28 days. Lumateperone acts primarily as a serotonin 5-HT2A receptor antagonist with high affinity, a postsynaptic dopamine D2 receptor antagonist with moderate affinity, an inhibitor of the serotonin reuptake transporter (SERT), and has moderate binding to dopamine D1 receptors—potentially contributing to indirect activation of NMDA and AMPA neurotransmission. The drug’s unique mechanism targets multiple neurotransmitter systems implicated in schizophrenia and related disorders[8][10]. As of now, lumateperone LAI is in clinical development and not yet approved for any indication.
