LUMC-BOB1-B7-TCR.1 is an investigational adoptive cellular immunotherapy consisting of autologous T cells engineered to express a high-affinity T-cell receptor (TCR) specific for the B-cell-specific octamer-binding protein 1 (BOB1), also known as POU2AF1. Developed by the Leiden University Medical Center (LUMC), this therapy targets a BOB1-derived peptide presented by the HLA-B*07:02 MHC class I molecule. BOB1 is a transcriptional coactivator essential for B-cell differentiation and is highly and consistently expressed across various B-cell malignancies, including B-cell acute lymphoblastic leukemia, multiple myeloma, and non-Hodgkin B-cell lymphoma, while being absent in non-hematopoietic tissues. This specificity allows the engineered T cells to selectively recognize and eliminate malignant B cells and healthy B cells (resulting in B-cell aplasia), while sparing other vital tissues. The therapy is currently being evaluated in early-phase clinical trials for patients with refractory B-cell malignancies.
