lumipodlin + osimertinib

lumipodlin + osimertinib (ABSK043 + Tagrisso) is an investigational, fully oral combination therapy being developed for the treatment of patients with EGFR-mutated, PD-L1-positive locally advanced or metastatic non-small cell lung cancer (NSCLC). The combination pairs lumipodlin (ABSK043), Abbisko Therapeutics' potentially first-in-class oral small-molecule PD-L1 inhibitor, with osimertinib (Tagrisso), AstraZeneca's approved third-generation, irreversible EGFR tyrosine kinase inhibitor (TKI). Lumipodlin binds to PD-L1 and induces its internalization from the cell surface, thereby blocking the PD-1/PD-L1 interaction and relieving PD-L1-mediated T-cell suppression. Osimertinib selectively inhibits EGFR-mutant tyrosine kinases. This combination aims to address the clinical challenge of EGFR-mutant NSCLC patients with high PD-L1 expression, who typically derive less benefit from EGFR-TKI monotherapy.
