ABSK043 + osimertinib is a combination therapy pairing lumipodlin (ABSK043), a first-in-class orally bioavailable small-molecule PD-L1 inhibitor, with osimertinib, a third-generation irreversible EGFR tyrosine kinase inhibitor. Developed by Abbisko Therapeutics in collaboration with AstraZeneca, this combination is designed to treat patients with EGFR-mutated and PD-L1 positive locally advanced or metastatic non-small cell lung cancer (NSCLC). Lumipodlin works by selectively binding to and inducing the internalization of the PD-L1 receptor, thereby blocking the PD-1/PD-L1 interaction and restoring T-cell-mediated anti-tumor immunity. Osimertinib targets the mutated EGFR signaling pathway. The combination is currently being evaluated in Phase I/II clinical trials to address populations that derive limited benefit from EGFR-TKI monotherapy.
