Lunasin is a naturally occurring peptide first isolated from soybeans, also found in barley and rye. [4, 15] It was investigated as a potential treatment for Amyotrophic Lateral Sclerosis (ALS) based on a media report of a single patient experiencing a dramatic symptom reversal and preclinical data suggesting it could alter histone acetylation patterns, which are abnormal in ALS patients. [2] However, a subsequent 12-month, open-label clinical trial (NCT02709330) in 50 patients with ALS found no evidence that lunasin slowed, stopped, or reversed disease progression. [2, 5, 8] The study also failed to demonstrate a significant effect on the proposed mechanism of histone H3 or H4 acetylation in blood samples. [2, 8] The treatment was associated with gastrointestinal side effects, most commonly constipation, and two serious cases of obstipation/fecal impaction requiring hospitalization were possibly related to the treatment. [2, 5] Lunasin has also been researched for its potential anti-inflammatory, antioxidant, and chemopreventive properties. [1, 3, 10]
