Lung Spheroid Stem Cells (LSCs) are a cellular therapeutic derived from lung tissue through a sequential 2D-3D-2D culture process. They are created by culturing outgrowth cells from lung tissue explants, allowing them to self-aggregate into three-dimensional spheroids, and then replating these spheroids onto fibronectin-coated surfaces to generate adherent lung spheroid cells. LSCs represent a natural mixture of lung progenitor cells (including Club Cells and AT2 cells) and supporting stromal cells without requiring antigenic sorting. These cells have shown therapeutic potential in treating pulmonary fibrosis by inhibiting apoptosis, fibrosis, and infiltration while promoting angiogenesis. LSCs can be derived from minimally invasive transbronchial biopsies, making them clinically feasible. When administered intravenously, they preferentially migrate to the lungs.
