Overview

**Luspatercept + deferasirox** is an investigational **combination therapy** being studied primarily for the treatment of transfusion-dependent anemia, such as in nonsevere aplastic anemia and beta-thalassemia. **Luspatercept** is a recombinant fusion protein that acts as an erythroid maturation agent, increasing late-stage erythropoiesis by acting as a ligand trap for transforming growth factor-beta (TGF-β) superfamily ligands, which enhances red blood cell production. **Deferasirox** is an oral iron chelator that binds excess iron, allowing it to be excreted and thereby managing iron overload due to chronic blood transfusions. The combination aims to address both the insufficient red cell production (via luspatercept) and iron overload (via deferasirox) in patients with transfusion-dependent anemias. This therapeutic strategy is under clinical investigation to assess its efficacy in improving hemoglobin levels and managing transfusional iron overload[2][4].
