Lutetium-177-DOTA-cetuximab (¹⁷⁷Lu-DOTA-cetuximab) is an investigational radioimmunotherapy (RIT) agent consisting of cetuximab, a chimeric IgG1 monoclonal antibody targeting the epidermal growth factor receptor (EGFR), conjugated to the macrocyclic chelator DOTA and radiolabeled with the beta-emitting radionuclide lutetium-177. By binding to EGFR, which is frequently overexpressed in head and neck squamous cell carcinoma (HNSCC), the agent delivers targeted cytotoxic beta radiation directly to tumor cells. It is being evaluated in a Phase 0/1 pilot theranostic clinical study (The ReCHANCE Study) led by Dr. Timothy Marr at Fremantle Hospital for patients with advanced HNSCC who have exhausted standard surgical and radiotherapy options.
