Overview

Lutetium Lu-177 anti-PD-1 is an experimental radioimmunotherapy (RIT) agent consisting of the beta-emitting radioisotope Lutetium-177 conjugated to a monoclonal antibody targeting Programmed Cell Death Protein 1 (PD-1). While PD-1 inhibitors are traditionally used in oncology to block inhibitory signals and activate T cells, this radiopharmaceutical construct is designed to utilize the PD-1 receptor as a docking site to deliver targeted cytotoxic radiation directly to activated T lymphocytes. In the context of autoimmune diseases like multiple sclerosis, the goal is to selectively deplete the pathogenic, recently activated T cells that express PD-1, thereby reducing neuroinflammation and disease progression. Preclinical studies have demonstrated synergistic effects when combined with other immunomodulators like fingolimod.
