Drug intelligence / Profile preview
Lutetium Lu 177 anti-TRA-1-60 (also known as [177Lu]Lu-CHX-A"-DTPA-Bstrongomab or [177Lu]Lu-Bstrongomab) is a targeted radiopharmaceutical therapy (RPT) being developed by Curemeta in collaboration with Wayne State University and the Barbara Ann Karmanos Cancer Institute. It consists of the humanized or chimeric monoclonal antibody Bstrongomab (or its scFv-Fc fragment), which targets the cancer-associated glycoepitope TRA-1-60 (TRA) expressed on podocalyxin (PODXL), conjugated to the beta-emitting radionuclide lutetium-177 (Lu-177). TRA-1-60 is a marker of pluripotency and cancer stemness that is highly expressed in aggressive, metastatic, and treatment-resistant cancers—such as pancreatic ductal adenocarcinoma (PDAC), prostate cancer, and ovarian cancer—while being minimally expressed in normal differentiated tissues. Upon binding to TRA-1-60 on the surface of cancer stem cells, the radiopharmaceutical is internalized and delivers localized beta radiation, inducing single- and double-strand DNA breaks that lead to tumor cell death. Preclinical studies have demonstrated significant tumor growth delay and antitumor efficacy, particularly when combined with DNA damage response (DDR) inhibitors like ATR or PARP inhibitors in ATM-deficient models.
