Luvomeitinib (FCN-159) is a potent, highly selective, and orally bioavailable small molecule inhibitor of mitogen-activated protein kinase kinase 1 and 2 (MEK1/2). By targeting these kinases, luvomeitinib effectively blocks the MAPK/ERK signaling pathway, which is a critical driver of cell proliferation and survival often dysregulated in various malignancies and genetic disorders. The drug was developed by Fosun Pharma (specifically its subsidiary Fosun Orinove) and has demonstrated significant clinical activity in histiocytic neoplasms, such as Langerhans cell histiocytosis (LCH) and Erdheim-Chester disease, as well as in neurofibromatosis type 1 (NF1) associated plexiform neurofibromas. In China, luvomeitinib has received Breakthrough Therapy Designation for the treatment of NF1-related plexiform neurofibromas in both pediatric and adult patients.
