Overview

LV-CD154 is an investigational ex vivo gene therapy consisting of a lentiviral vector encoding the human CD154 (CD40 ligand) protein. Developed primarily for the treatment of B-cell malignancies such as chronic lymphocytic leukemia (CLL), the therapy involves harvesting autologous tumor cells from a patient and transducing them with the lentiviral vector to induce surface expression of CD154. When these modified cells are re-infused, the expressed CD154 binds to the CD40 receptor on neighboring leukemic B-cells and other antigen-presenting cells. This interaction triggers the upregulation of costimulatory molecules (e.g., CD80, CD86) and the secretion of pro-inflammatory cytokines, effectively transforming the immunosuppressive tumor cells into potent stimulators of the immune system. This process bypasses the tumor's immune evasion mechanisms and elicits a systemic, T-cell-mediated anti-tumor immune response.
