LV3-siNlrp3 is a preclinical research tool designed to investigate the role of the NLRP3 inflammasome in the gut-liver-brain axis, particularly in the context of Parkinson's disease. It consists of a small interfering RNA (siRNA) sequence targeting the Nlrp3 gene, packaged within a lentiviral vector (LV3). In animal models, systemic administration via tail vein injection allows for the knockdown of NLRP3 expression in the liver. This intervention has been shown to reduce peripheral inflammation, suppress the production of pro-inflammatory cytokines, and subsequently mitigate neuroinflammation and dopaminergic neuron loss in the brain. It is primarily used in academic research to explore how peripheral immune responses contribute to neurodegenerative processes.
