Overview

LV5FU2 is a combination chemotherapy regimen consisting of leucovorin (folinic acid) and 5-fluorouracil (5-FU). In the context of the DOVIPA clinical trial (NCT05811130) sponsored by Hopital Foch, it is utilized as a maintenance treatment for patients with metastatic pancreatic adenocarcinoma following induction therapy with mFOLFIRINOX, dostarlimab, and vitamin D3. 5-fluorouracil is a pyrimidine analog that acts as an antimetabolite, primarily inhibiting thymidylate synthase to disrupt DNA synthesis and repair. Leucovorin is a reduced form of folic acid that acts as a biochemical modulator, enhancing the binding of 5-FU to thymidylate synthase and thereby increasing its cytotoxic efficacy. This regimen is a standard component of various gastrointestinal cancer treatments and is being evaluated in this specific combination to improve outcomes in Stage IV pancreatic cancer.
