Overview

LVK041 is an investigational, engineered Natural Killer (NK) cell therapy developed by Laverock Therapeutics for the treatment of solid tumors. The therapy utilizes Laverock's proprietary Gene Editing induced Gene Silencing (GEiGS) technology, which enables stable and programmable gene silencing by using gene editing tools (such as CRISPR/Cas9) to redirect endogenous RNA interference (RNAi) pathways toward specific target genes. In LVK041, this platform is applied to silence the Cytokine Inducible SH2-containing protein (CISH), an intracellular checkpoint that negatively regulates NK cell activation, metabolism, and effector functions. By silencing CISH, LVK041 is designed to enhance the persistence, metabolic fitness, and anti-tumor cytotoxicity of NK cells within the immunosuppressive tumor microenvironment, potentially providing a more effective treatment for various solid malignancies.
