LVP-R-G-mIL2 is an experimental recombinant oncolytic vaccinia virus (Lister strain) engineered for cancer immunotherapy. It is designed to co-express murine Interleukin-2 (mIL2), the NKG2D ligand RAE-1γ (represented by 'R'), and the tumor-associated antigen gp70 (represented by 'G'). The therapeutic strategy utilizes the virus's inherent oncolytic properties to directly lyse tumor cells while simultaneously delivering immune-modulating payloads. The expression of mIL2 stimulates T-cell and NK-cell proliferation and activity, while RAE-1γ enhances innate immune recognition via the NKG2D receptor. The gp70 antigen provides a specific target for the induction of adaptive anti-tumor immunity. This multi-pronged approach is intended to overcome the immunosuppressive tumor microenvironment, particularly in models of mammary carcinoma such as the 4T1 breast cancer line.
