LVP-R-G-SPARC + gp70 peptides + murine interleukin-2 is an experimental combination immunotherapy regimen investigated for the treatment of murine mammary carcinoma, specifically in the 4T1 breast cancer model. The regimen centers on a recombinant oncolytic vaccinia virus (LVP-R-G-SPARC) derived from the Lister strain, which is engineered for tumor-selective replication and direct oncolysis. This virus construct typically incorporates red and green fluorescent proteins (RFP/GFP) for tracking and is designed to target or express SPARC (Secreted Protein Acidic and Rich in Cysteine), a protein frequently overexpressed in the tumor microenvironment. The regimen is augmented by the administration of gp70-derived peptides, which serve as tumor-associated antigens to prime a specific T-cell response, and murine interleukin-2 (mIL2), a cytokine that acts as an adjuvant to promote the expansion and activation of cytotoxic T lymphocytes and natural killer cells. This multi-modal approach combines direct viral destruction of the tumor with a robust, antigen-specific systemic immune response to overcome tumor-induced immunosuppression.
