Overview

LW1646 is a newly synthesized, highly specific, and potent phosphodiesterase 5 (PDE5) inhibitor. Developed by researchers at Sun Yat-sen University and Hainan University, LW1646 is being investigated for its therapeutic potential in chronic kidney disease, specifically in ameliorating renal fibrosis and lipid-induced kidney injuries. Mechanistically, LW1646 activates the cGMP/PKG signaling pathway, which alleviates endoplasmic reticulum (ER) stress, maintains mitochondrial homeostasis, and inhibits the formation of mitochondria-associated membranes (MAMs), thereby reducing mitochondrial calcium overload and preventing the progression of kidney fibrosis.
