Overview

LX1033 is an orally administered, small-molecule inhibitor of tryptophan hydroxylase, the rate-limiting enzyme in serotonin synthesis. It was developed by Lexicon Pharmaceuticals for the treatment of diarrhea-predominant irritable bowel syndrome (IBS-D). The drug is designed to act locally within the gastrointestinal tract to reduce serotonin production, thereby modulating gut motility and secretion without affecting central nervous system serotonin levels. Clinical trials demonstrated that LX1033 significantly reduced plasma and urinary 5-HIAA (a biomarker for serotonin synthesis) and showed some positive effects on abdominal pain in IBS-D patients. However, development was discontinued after Phase II trials due to insufficient efficacy over placebo in primary endpoints[1][2][4][6].
