Overview

LX4211 + metformin is a combination therapy consisting of **LX4211 (sotagliflozin)**, a dual inhibitor of sodium-glucose cotransporter 1 (SGLT1) and sodium-glucose cotransporter 2 (SGLT2), and **metformin**, a standard first-line oral antihyperglycemic agent. LX4211 blocks SGLT2 in the kidneys to increase urinary glucose excretion and blocks SGLT1 in the intestine to delay and reduce glucose absorption, thereby reducing postprandial glucose excursions. This dual inhibition also increases glucagon-like peptide 1 (GLP-1) and peptide YY (PYY) secretion, both of which have beneficial metabolic effects. Metformin primarily works by decreasing hepatic glucose production and improving insulin sensitivity. The combination is being studied for improved glycemic control in patients with type 2 diabetes mellitus (T2DM) inadequately controlled with metformin monotherapy, with a focus on hemoglobin A1c reduction, body weight, and cardiovascular risk factors[1].
