LY01610 is an intravenously administered, polyethylene glycolated liposomal formulation of irinotecan hydrochloride being developed by Luye Pharma for the treatment of solid tumors, notably relapsed small cell lung cancer (SCLC) and advanced esophageal squamous cell carcinoma (ESCC).[1][3][13] By encapsulating irinotecan in PEGylated liposomes, LY01610 alters the pharmacokinetics and biodistribution of irinotecan and its active metabolite SN-38, increasing and prolonging systemic exposure, enriching drug concentrations in tumor tissue, and reducing exposure in normal organs, with the aim of enhancing antitumor efficacy and reducing toxicity relative to conventional irinotecan.[1][3][5][7] Clinically, LY01610 has shown promising antitumor activity and manageable safety in Phase I–II studies in ESCC and relapsed SCLC, and is in a randomized Phase III trial versus topotecan as second-line therapy for recurrent SCLC.[1][2][3][4][11]
