LY2090314 + gemcitabine is a combination regimen comprising LY2090314, a potent and selective small molecule inhibitor of glycogen synthase kinase-3 beta (GSK-3β), and gemcitabine, a nucleoside analog and antineoplastic agent commonly used in various solid tumors including pancreatic cancer. LY2090314 inhibits both GSK-3α and GSK-3β isoforms, activating Wnt/β-catenin signaling, stabilizing β-catenin, and promoting apoptosis in cancer cells. Gemcitabine works by incorporating into DNA and inhibiting DNA synthesis, causing cell death. The combination was investigated in clinical trials for metastatic pancreatic cancer and has been evaluated for potential synergistic antitumor activity in preclinical and early clinical studies[1][4][5].
