Overview

LY2275796 is a second-generation antisense oligonucleotide designed to inhibit the expression of eukaryotic translation initiation factor 4E (eIF-4E), a protein that regulates mRNA translation and is upregulated in various cancers. By blocking eIF-4E, LY2275796 reduces the synthesis of tumor angiogenic factors and growth/survival factors, thereby inhibiting tumor progression, angiogenesis, and metastasis. The drug was developed through a collaboration between Isis Pharmaceuticals (now Ionis Pharmaceuticals) and Eli Lilly. It has been investigated primarily for advanced solid tumors in clinical trials but has not demonstrated significant tumor response as monotherapy[1][2][3][4][5][7].
