LY2510924 is a potent and selective cyclic peptide antagonist of the CXC chemokine receptor 4 (CXCR4). It blocks the binding of stromal cell-derived factor-1 (SDF-1, also known as CXCL12) to CXCR4, thereby inhibiting downstream receptor activation and signaling. This mechanism disrupts SDF-1/CXCR4-mediated processes such as tumor cell migration, proliferation, survival signals, and metastasis. LY2510924 has demonstrated antitumor activity in preclinical models of solid tumors and hematologic malignancies including acute myeloid leukemia (AML), non-Hodgkin lymphoma, renal cell carcinoma, lung cancer, colon cancer, and breast cancer. In clinical studies—including phase 1 and phase 2 trials—LY2510924 has been evaluated both as monotherapy and in combination with other agents for various cancers.
