LY2510924 + durvalumab is an experimental combination therapy for cancer, consisting of LY2510924, a selective peptide antagonist of the chemokine receptor CXCR4, and durvalumab, a human monoclonal antibody targeting programmed death-ligand 1 (PD-L1). LY2510924 blocks the interaction of CXCR4 (expressed on tumor and hematopoietic cells) with its ligand SDF-1, thus inhibiting tumor cell migration, metastasis, and mobilizing immune cells. Durvalumab inhibits PD-L1 to restore antitumor immune response. The combination has been investigated in phase 1 trials for advanced refractory solid tumors, where it showed an acceptable safety profile and some evidence of antitumor activity, with the recommended phase 2 dose being LY2510924 40 mg subcutaneously once daily and durvalumab 1500 mg intravenously every 28 days[1][3].
