LY2584702 is an orally available, small molecule inhibitor of ribosomal protein S6 kinase (p70S6K), a serine/threonine kinase that acts downstream in the phosphatidylinositol 3‑kinase (PI3K) pathway. By competitively inhibiting ATP binding to p70S6K, it prevents phosphorylation of the S6 subunit of ribosomes, leading to reduced protein synthesis and cellular proliferation in tumor cells. This mechanism targets a pathway often upregulated in various cancers and involved in cell growth, proliferation, motility, and survival. Developed by Eli Lilly for oncology indications including advanced cancer and renal cell carcinoma, clinical trials have shown limited efficacy as monotherapy or in combination with other agents[1][3][4].
