Overview

LY2584702 + erlotinib is an investigational combination regimen consisting of LY2584702, a selective oral ATP-competitive inhibitor of p70 S6 kinase (p70S6K), and erlotinib, an oral reversible EGFR (epidermal growth factor receptor) tyrosine kinase inhibitor. The rationale for the combination is to simultaneously inhibit the EGFR pathway and downstream signaling via the S6 kinase pathway, both implicated in cellular proliferation and cancer survival signaling. Preclinical and early clinical studies have assessed this combination for synergistic anticancer effects, particularly in solid tumors such as head and neck squamous cell carcinoma (HNSCC) and non-small cell lung cancer (NSCLC). However, while in vitro models showed enhanced cell death, clinical experience was limited by significant toxicity and tolerability challenges.
