Overview

LY2624587 is a fully humanized monoclonal antibody that acts as a potent and selective antagonist of the chemokine receptor CXCR4. It blocks the binding of stromal cell-derived factor 1 (SDF-1, also known as CXCL12) to CXCR4, thereby inhibiting SDF-1-induced signaling pathways such as MAPK and AKT activation. This results in receptor internalization, down-regulation of surface CXCR4, inhibition of tumor cell migration, and induction of apoptosis in hematologic cancer cells both in vitro and in vivo. LY2624587 was developed for the treatment of hematological malignancies including acute myeloid leukemia, chronic lymphocytic leukemia, and non-Hodgkin's lymphoma. The drug demonstrated dose-dependent tumor growth inhibition and survival benefit in preclinical models and advanced into clinical studies for cancer.
