Overview

LY2811376 is an orally available, non-peptidic small molecule inhibitor of β-site amyloid precursor protein cleaving enzyme 1 (BACE1), developed by Eli Lilly. It selectively inhibits BACE1 over BACE2 and other aspartic proteases, with an IC50 of approximately 239–249 nM for BACE1 and demonstrates robust reduction of amyloid-β (Aβ) levels in preclinical models and humans[1][3][4]. The drug was designed to target the amyloidogenic processing pathway implicated in Alzheimer's disease by reducing Aβ production through inhibition of BACE1-mediated cleavage of APP[4][7]. Despite promising biomarker changes and central nervous system penetration in early clinical studies, development was discontinued due to toxicology findings unrelated to its mechanism[4].
