LY2879788 is a small molecule antagonist of the metabotropic glutamate receptors 2 and 3 (mGluR2/3), which was developed by Eli Lilly for the treatment of major depressive disorder (MDD). By blocking these presynaptic receptors, LY2879788 prevents the negative feedback inhibition of glutamate release, thereby increasing synaptic glutamate levels and potentially exerting rapid-acting antidepressant effects similar to those observed with NMDA receptor antagonists like ketamine. Although it reached Phase 2 clinical testing, the development of LY2879788 was ultimately discontinued, and it is no longer listed in the company's active pipeline.
