Overview

LY2881835 is a potent and selective small molecule agonist of the G protein-coupled receptor 40 (GPR40), also known as free fatty acid receptor 1 (FFAR1). It was developed as an oral antihyperglycemic agent for the treatment of type 2 diabetes mellitus. The drug acts by stimulating GPR40, which enhances glucose-dependent insulin secretion from pancreatic beta cells and increases GLP-1 secretion, leading to improved glycemic control. Preclinical studies demonstrated that LY2881835 provides immediate and durable reductions in blood glucose levels in rodent models of both early- and late-stage type 2 diabetes. Clinical development reached phase I trials but was discontinued for type 2 diabetes[1][4][7].
