Overview

LY2940094 (also known as BTRX-246040) is a potent, selective, and orally available small molecule antagonist of the nociceptin receptor (NOP receptor). It was developed by Eli Lilly and later BlackThorn Therapeutics for neurobehavioral disorders, primarily major depressive disorder (MDD), where it demonstrated proof-of-concept clinical efficacy. The drug has also been investigated for alcohol dependence and Parkinson's disease. Mechanistically, it acts by blocking the NOP receptor with high affinity (\(K_i = 0.105\) nM), reducing ethanol self-administration in animal models and showing antidepressant-like effects in preclinical studies. Clinical development reached phase II trials for MDD and alcohol dependence; development for alcoholism was discontinued[1][2][3][6][7].
