LY2969822 is an oral prodrug developed by Eli Lilly that is rapidly converted in the body to LY2934747, a potent and selective agonist of metabotropic glutamate 2/3 (mGlu2/3) receptors. It was developed for the potential treatment of chronic pain conditions. Preclinical studies demonstrated that activation of mGlu2/3 receptors by this compound attenuates pain behaviors across a broad range of rodent models, including neuropathic, inflammatory, and visceral pain. The analgesic effects are mediated through suppression of spinal cord neuron activity associated with pain signaling. The drug was investigated in Phase 1 clinical trials for safety and pharmacokinetics in healthy volunteers but has not advanced to later-stage clinical development[1][4][5][6].
