LY3009120 is an orally available, synthetic small molecule that acts as a pan-inhibitor of the serine/threonine protein kinase Raf family, targeting A-Raf, B-Raf (including BRAFV600E and BRAFWT), and C-Raf with nanomolar potency. It inhibits RAF-mediated signal transduction pathways, particularly the RAF/MEK/ERK signaling cascade, which is frequently dysregulated in human cancers and plays a critical role in tumor cell proliferation and survival. Preclinical studies have demonstrated that LY3009120 induces anti-proliferative effects through G1 cell cycle arrest and apoptosis in cancer cell lines harboring BRAF or KRAS mutations. The compound has also shown anti-adipogenic effects by modulating key transcription factors involved in adipocyte differentiation. Developed for its potential antineoplastic activity, clinical development reached Phase 1 trials across multiple indications.
