LY3023414 + necitumumab is an experimental combination therapy comprised of LY3023414, a potent oral small molecule inhibitor of class I PI3K isoforms, mTOR, and DNA-PK, together with necitumumab, a fully human IgG1 monoclonal antibody targeting the extracellular domain III of the epidermal growth factor receptor (EGFR). LY3023414 inhibits PI3K/mTOR signaling, thereby affecting tumor cell proliferation and survival, while necitumumab blocks ligand binding to EGFR, prevents receptor dimerization and signaling, and induces antibody-dependent cellular cytotoxicity (ADCC). The combination was studied in squamous non-small cell lung cancer, but a Phase I/II trial (NCT02443337) was terminated due to lack of efficacy.
