Overview

LY3031207 is a novel small molecule inhibitor of microsomal prostaglandin E synthase 1 (mPGES-1), developed by Eli Lilly. It was investigated as an orally administered anti-inflammatory agent. The drug underwent phase I clinical trials in healthy volunteers to assess safety, tolerability, pharmacokinetics, and pharmacodynamics. However, development was terminated after multiple cases of dose-dependent drug-induced liver injury (DILI) with hypersensitivity features were observed during the trial. The mechanism of hepatotoxicity may involve metabolic activation of the imidazole ring present in its structure[2][5][3].
