Overview

LY303511 (often referred to as LY30 in specific research contexts) is a small molecule pyranone derivative and a structural analogue of the well-known PI3K inhibitor LY294002. Notably, LY303511 lacks the ability to inhibit PI3K, which has led to its widespread use as a negative control in signaling studies. However, research has demonstrated that LY303511 possesses independent biological activity, specifically acting as a potent sensitizer of cancer cells to Tumor Necrosis Factor-Related Apoptosis-Inducing Ligand (TRAIL)-mediated apoptosis. Its mechanism of action involves the upregulation and clustering of death receptors (DR4 and DR5), the downregulation of the anti-apoptotic protein c-FLIP, and the induction of intracellular reactive oxygen species (ROS), particularly superoxide. These effects lead to the synergistic activation of the extrinsic apoptotic pathway in various cancer cell lines, including cervical cancer (HeLa) and other solid tumors. It was primarily developed as a research tool by Eli Lilly and extensively studied for its anti-cancer properties by researchers at the National University of Singapore.
