Overview

LY3056480 is a small molecule gamma-secretase inhibitor developed for the treatment of sensorineural hearing loss. Its mechanism of action involves inhibition of Notch signaling, which is believed to promote regeneration of inner ear sensory hair cells lost due to aging or damage. Preclinical studies demonstrated that pharmacological inhibition of Notch signaling with LY3056480 can induce supporting cells in the cochlea to transdifferentiate into new hair cells, leading to partial recovery of hearing in animal models. The drug is administered locally via intratympanic injection, targeting the inner ear while minimizing systemic exposure. Clinical trials (Phase I/IIa) have shown that LY3056480 is safe and well tolerated in adults with mild-to-moderate sensorineural hearing loss; although primary efficacy endpoints were not met, post-hoc analyses indicated signs of clinical activity in some patients[1][2][3][5][6][7].
