Overview

LY3143753 is a long-acting fibroblast growth factor 21 (FGF21) analog developed by Eli Lilly for the treatment of metabolic disorders, including non-alcoholic steatohepatitis (NASH). As an FGF21 mimetic, it targets the FGFR1c/beta-Klotho receptor complex to modulate lipid and glucose metabolism, enhance insulin sensitivity, and reduce hepatic fat accumulation. The molecule was engineered for an extended half-life to support a less frequent dosing regimen compared to native FGF21. While it entered Phase 1 clinical testing to evaluate safety and pharmacokinetics in healthy subjects, Eli Lilly discontinued the development of LY3143753 in 2021.
