Overview

LY3154885 is an orally active, synthetic small molecule that functions as a positive allosteric modulator (PAM) of the human dopamine D1 receptor. It was developed to have similar pharmacologic properties to mevidalen, another D1 PAM, but with a reduced risk of drug-drug interactions due to its primary metabolism by UGT enzymes rather than CYP3A4. LY3154885 was investigated as a clinical candidate for neurological disorders such as Lewy body dementia and other conditions where modulation of dopamine signaling may be beneficial. The drug entered Phase 1 clinical trials in 2019 but development was terminated in early 2020 for business reasons[1][3][4][5][7].
