Overview

This is a combination regimen including LY3214996, nab-paclitaxel (nanoparticle albumin-bound paclitaxel), and gemcitabine. **LY3214996** is an oral small molecule inhibitor targeting ERK1/2 kinases in the MAPK pathway, developed for tumors characterized by alterations in genes such as KRAS, BRAF, NF1, and MAP2K1/2. **Nab-paclitaxel** is a nanoparticle albumin-bound formulation of the microtubule inhibitor paclitaxel, which stabilizes microtubules and inhibits mitosis. **Gemcitabine** is a nucleoside analog (antimetabolite) that inhibits DNA synthesis. The triple combination is under investigation primarily for the treatment of metastatic pancreatic cancer and other solid tumors, especially those with MAPK pathway alterations or KRAS mutations. This regimen is not approved but is under clinical investigation.
