Overview

LY3300054 + abemaciclib is a combination of a monoclonal antibody immune checkpoint inhibitor, LY3300054 (an anti–PD-L1 antibody), with abemaciclib, an orally active small molecule inhibitor of cyclin-dependent kinases CDK4 and CDK6. LY3300054 blocks the interaction between PD-L1 and PD-1 to restore antitumor immune activity, while abemaciclib inhibits cancer cell proliferation by preventing phosphorylation of the retinoblastoma protein and inducing cell cycle arrest. This combination has been investigated in advanced, refractory solid tumors, including breast and pancreatic cancers, in early phase clinical trials. Primary indications studied include advanced solid tumors (general), with emphasis on cancers that may benefit both from immune checkpoint blockade and cell-cycle inhibition[1][5].
