Overview

LY3415244 is a bispecific monoclonal antibody designed to simultaneously target and inhibit both TIM-3 (T-cell immunoglobulin and mucin-domain containing-3) on immune cells, including tumor-infiltrating lymphocytes, and PD-L1 (programmed death-ligand 1) on tumor cells. By blocking these two immune checkpoint pathways, the drug was hypothesized to overcome primary and acquired resistance to anti-PD-(L)1 therapies in patients with advanced solid tumors. Developed by Eli Lilly, LY3415244 entered phase I clinical trials for the treatment of advanced solid tumors but demonstrated high immunogenicity with frequent development of antidrug antibodies (ADA), leading to reduced target engagement and hypersensitivity reactions such as anaphylaxis. Due to these safety concerns and early termination of the study, further development was discontinued[2][3][6].
