Overview

LY3475070 is an orally bioavailable, small molecule inhibitor of the ectoenzyme CD73 (cluster of differentiation 73; also known as 5'-ecto-nucleotidase or 5'-NT), developed by Eli Lilly for the treatment of advanced cancers. By binding to and inducing internalization of CD73, LY3475070 blocks the conversion of adenosine monophosphate (AMP) to adenosine in the tumor microenvironment. This reduces immunosuppressive adenosine levels, thereby enhancing anti-tumor immune responses by increasing activity of cytotoxic T cells and natural killer cells while reducing regulatory T cell and myeloid-derived suppressor cell function. The drug has been evaluated as monotherapy and in combination with pembrolizumab in Phase 1 clinical trials for multiple solid tumors including triple negative breast cancer, pancreatic cancer, non-small cell lung cancer, renal cell carcinoma, melanoma, prostate cancer, and ovarian cancer[1][2][4][5][7].
