Overview

LY3526318 is an orally administered, potent, and selective small molecule antagonist of the transient receptor potential ankyrin 1 (TRPA1) ion channel. It was developed by Eli Lilly as a non-opioid analgesic for the treatment of chronic pain conditions such as diabetic peripheral neuropathic pain (DPNP), chronic low back pain (CLBP), and osteoarthritis (OA) pain. The drug exhibits limited penetration into the central nervous system and acts primarily through peripheral TRPA1 inhibition. Clinical trials have shown that while it has an acceptable safety profile in phase 1 studies, efficacy results in phase 2 trials were mixed; no significant difference from placebo was observed in OA or DPNP populations, though some numerical improvement was seen in CLBP. A potential risk for drug-induced hepatotoxicity has been noted[1][4][5][7].
