Overview

LY-377604 is a small molecule drug developed as a human β3-adrenergic receptor agonist and β1- and β2-adrenergic receptor antagonist. It has no sympathomimetic activity at the β1- and β2-adrenergic receptors. The compound was investigated for its potential to treat obesity, particularly in combination with sibutramine (a norepinephrine-serotonin uptake inhibitor), where it was hypothesized to ameliorate side effects such as increased blood pressure and heart rate associated with sibutramine treatment. In preclinical studies, LY-377604 demonstrated the ability to enhance energy expenditure and fat oxidation while preserving lean body mass[1][5][7].
