Overview

LY3847429 is an orally bioavailable, small-molecule inhibitor of extracellular signal-regulated kinases 1 and 2 (ERK1/2), developed by Eli Lilly and Company for the treatment of advanced solid tumors. As a terminal component of the MAPK/ERK signaling pathway, ERK1/2 plays a critical role in regulating cell proliferation, differentiation, and survival. Many malignancies exhibit overactivation of this pathway due to upstream mutations in RAS or BRAF. By selectively inhibiting ERK1/2, LY3847429 aims to block the oncogenic signaling cascade that drives tumor growth, particularly in cases where resistance to BRAF or MEK inhibitors has developed. The drug has been evaluated in Phase 1 clinical trials for patients with advanced solid tumors, though it is no longer listed in Eli Lilly's active development pipeline.
