Overview

LY3962681 is a first-in-class, investigational small interfering RNA (siRNA) therapy designed to target and reduce the expression of alpha-synuclein (*SNCA*) mRNA, thereby lowering alpha-synuclein protein levels in the brain. It is administered intrathecally (into the spinal fluid) and is being developed for the treatment of Parkinson's disease. The drug aims to address the underlying pathology of Parkinson’s by reducing pathological alpha-synuclein accumulation, which has strong genetic and clinical links to disease progression. Preclinical studies have shown dose-dependent suppression of *SNCA* mRNA and protein in relevant brain regions, with good tolerability observed in toxicology studies. Clinical development is currently at Phase 1, focusing on safety, tolerability, pharmacokinetics/pharmacodynamics (PK/PD), and effects on cerebrospinal fluid alpha-synuclein levels[3][6][1].
