LY4152199 is a first-in-class bispecific antibody that targets both the B-cell activating factor receptor (BAFF-R, also known as TNFRSF13C) and CD3 on T cells. By binding to BAFF-R on malignant B cells and CD3 on T cells, it acts as a T-cell engager, redirecting cytotoxic T lymphocytes to eliminate BAFF-R–expressing B cell malignancies. Preclinical studies have shown that LY4152199 induces potent, concentration-dependent T-cell activation and cytotoxicity against malignant B cells with lower cytokine release compared to other bispecific antibodies such as mosunetuzumab. It is being developed by Eli Lilly for the treatment of relapsed or refractory B cell malignancies, including cases with loss of CD19/CD20 expression[1][3][5][6].
